2. What Is CJC-1295?
CJC-1295 is a synthetic analog of growth hormone-releasing hormone (GHRH), built on the first 29 amino acids of GHRH (the GRF 1-29 fragment) with substitutions that make it more stable. It is studied as a growth hormone-releasing factor in research models. Two forms appear in the literature and on the market.
CJC-1295 with DAC (Drug Affinity Complex) adds a chemical group that binds blood albumin to extend its presence in circulation, and carries CAS number 446262-90-4 with the molecular formula C165H269N47O46. CJC-1295 without DAC, often called Modified GRF 1-29, carries CAS number 863288-34-0 with the molecular formula C152H252N44O42. The practical difference is duration: the DAC version is designed for a much longer half-life in research models, while the no-DAC version acts over a shorter window. Both are supplied as lyophilized powders.
3. Research Background
CJC-1295 came out of work on GRF analogs in the mid-2000s, with the DAC technology designed to address the very short half-life of natural GHRH. A 2006 study by Teichman and colleagues in the Journal of Clinical Endocrinology and Metabolism reported prolonged stimulation of growth hormone and IGF-I secretion by CJC-1295 (PubMed 16352683), one of the few human-subject reports on the compound.
Much of the later literature is analytical and centered on anti-doping detection, since CJC-1295 is a prohibited substance in sport. Papers by Henninge and colleagues in 2010 (PubMed 21204297), Timms and colleagues in 2019 (PubMed 30489688), and Memdouh and colleagues in 2021 (PubMed 34665524) in Drug Testing and Analysis describe methods for detecting CJC-1295 and related GHRH analogs.
Outside that 2006 report, controlled human efficacy data is sparse, and most mechanistic understanding comes from the broader GHRH literature.
4. Mechanism of Action
CJC-1295 works through the GHRH receptor on the pituitary gland. By mimicking GHRH, it is studied for its ability to prompt the pituitary to release growth hormone in pulses, which in turn raises IGF-I. The substitutions in the sequence protect it from the enzyme (DPP-4) that rapidly degrades natural GHRH, and the DAC version adds albumin binding on top of that for a longer effect.
Because it acts upstream at the GHRH receptor, CJC-1295 is mechanistically different from ghrelin-receptor secretagogues such as ipamorelin. The two are often studied together precisely because they act on different receptors, and combination research formats reflect that pairing.
Researchers should note the substantial anti-doping detection literature around this compound and the limited controlled efficacy data beyond the early human report. For current work, search PubMed directly.
5. Storage and Stability
Lyophilized CJC-1295 is stored at -20°C for long-term holding, shielded from light, and repeated freeze-thaw cycles are avoided. Stability for a given lot is documented on its Certificate of Analysis. Refrigeration at 2°C to 8°C suits shorter windows, with the vial protected from light and moisture.
Freeze-thaw cycling is avoided. Because the DAC and no-DAC forms differ, the lot-specific Certificate of Analysis should be checked for the exact material on hand.
